The most frequently mutated gene among all genes known to be involved in human cancers is the tumor suppressor p53. As its product can activate transcription of numerous downstream genes, p53 is likely to mediate a variety of biological functions through transcriptional regulation of those targets. p21Waf1 and BAX are key mediators of two major p53 functions: cell cycle arrest and apoptosis. Although these functions had been considered the essential features of p53-dependent tumor suppression, our recent identification of p53R2, a p53 target that supplies deoxyribonucleotides for repairing DNA, proved that p53 is directly involved in the DNA repair process as well. |
